欧石楠体细胞胚再生过程中遗传稳定性的RAPD和ISSR分析

摘 要:

利用RAPD和ISSR分子标记, 研究了欧石楠体细胞胚再生过程中4个不同发育时期的遗传稳定性, 结果表明, 2种分子标记方法均能揭示材料间较高的遗传稳定性。从190对引物组合中筛选出9对适宜的RAPD扩增引物, 共扩增获得158条带, 平均每对引物可扩增出17.56条带, 其中多态性条带13条, 比例为8.23%; 材料间遗传相似系数的变化幅度为0.8403~0.9625, 平均值为0.8979。从45对引物组合中筛选出8对适宜的ISSR扩增引物, 共扩增获得143条带, 平均每对引物可扩增出17.875条带, 其中多态性条带15条, 比例为10.48%, 材料间遗传相似系数变化范围为0.8154~0.9032, 平均值为0.8618。聚类分析结果表明, 2种分子标记均能将供试材料分开, 且具有一定的相似性, 但也存在差异。

Genetic Stability Analysis in Somatic Embryo Regeneration of Erica carnea by RAPD and ISSR Markers

Abstract:

Genetic diversity in Erica carnea field seedlings, embryogenic calli, adventitious buds and regenerated seedlings was analyzed by random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) makers. The results showed that there was a high level of genetic stability. In RAPD analysis, 9 primers screened from 190 RAPD primers were polymorphic. A total of 158 bands were observed in 9 primers with an average of 17.56 bands, among which 13 bands were polymorphic and the percentage of polymorphic bands (PPB) was 8.23%. The RAPD-based genetic similarity (RAPD-GS) among the material ranged from 0.8403 to 0.9625 with an average of 0.8979. In ISSR analysis, 8 primers screened from 45 RAPD primers were polymorphic. A total of 143 bands were detected in 8 primers, with an average of 17.875, among which 15 bands were polymorphic and PPB was 10.48%. The ISSR-derived genetic similarity (ISSR-GS) ranged from 0.8154 to 0.9032 with an average of 0.8618. The cluster analysis indicated that the tested materials could be distinguished by both RAPD and ISSR markers.
